Research Statement

Increasingly more neurodegenerative diseases are showing a mis-sorting of synaptic vesicles. Therefore it is necessary to understand how synaptic vesicles traffic properly from the cell body to the synapse. It has been previously demonstrated by our lab that the adaptor protein AP-3 regulates the sorting of synaptic vesicles. My project to determine what cargo are regulated by AP-3 in neurons and how those cargo are mis-trafficked in the absence of binding to AP-3. To do this, we will examine known cargo of AP-3 in fixed primary cultured neurons, live-imaging of tagged cargo, EM of brain sections.
